要辣椒是茄科辣椒属植物,为一年或多年生草本植物,具有药食同源的特点,主要作为烹饪用香辛料应用在日常生活中。近年来还把辣椒作为保健类物质的提取原料来使用,如:从辣椒中提取辣椒碱和辣椒红色素等。目前国内外已有许多关于从植物中提纯黄酮类化合物的研究,但对于辣椒中黄酮类化合物的提取、纯化和利用则鲜有报道。因此,本文以辣椒为研究对象,主要对辣椒中黄酮类化合物的提纯条件进行探索,并对其纯化工艺进行设计。通过对辣椒中黄酮类化合物的纯化工艺的设计,为开发利用我国的辣椒黄酮资源提供科学依据,同时也为黄酮类化合物的工业化生产以及其在食品工业领域的开发和应用提供理论依据。本文的主要研究内容及结果如下:(1)采用微波辅助提取法对辣椒果皮中的黄酮类化合物进行提取,提取条件为:乙醇浓度 35%、料液比 1:55、微波时间 4min、微波功率 119W。得到的辣椒果皮中黄酮类化合物的提取率为 8.63mg/g。(2)选择 NKA-9 型树脂作为纯化辣椒皮黄酮的理想树脂,并对该树脂的纯化工艺参数进行了研究,结果表明在使用 NKA-9 型树脂纯化辣椒中黄酮类化合物时,其最佳吸附条件为:上样吸附速率为 0.59mL/min、上样液 pH值 5、上样量 20mL,此时的辣椒黄酮吸附率可达到 72.03%。最佳的洗脱条件为:洗脱速率为 1mL/min、用浓度为 70%的乙醇溶液作为洗脱剂、洗脱剂用量为 50mL,此时的辣椒黄酮洗脱率可达到 92.38%。关键词 辣椒 大孔吸附树脂 柱层析 黄酮类化合物 纯化 吉林大学学士学位毕业论文0ABSTRACTCapsicum annuum is capsicum of Solanaceae,which is annual or perennial herb.It can be used as food and medicine at the same time and mainly as cooking spices applications in daily life.In recent years,Capsicum annuum also used as raw materials for the extraction of health substances.For example:Capsaicin and Paprika Oleoresin.Presently there has been many research of the purification of flavonoids from plants at home and abroad.But for the extraction, purification and utilization of flavonoids from Capsicum annuum are rarely reported.Therefore, this article use Capsicum annuum as the research object,mainly explore the purification conditions of the flavonoids compound in Capsicum annuum and design the purification process.By the designing of purification process of Chili flavonoids,provide a scientific basis for the development and utilization of Chili flavonoids resources in our country,also provide a theoretical basis for flavonoids industrialized production as well as the development and application in the food industry .The main research contents and results are as follows:(1)Using microwave assisted extraction method extract the flavonoids in Capsicum annuum skin.The parameters for extraction are:ethanol concentration 35%,ratio of solid to liquid 1:55 and microwave power at 119W for 4min.The extraction rate of flavonoids from Capsicum annuum skin is 8.63mg/g.(2)Choose NKA-9 resin as the ideal resin for the purification of Capsicum annuum skin flavonoids and the resin purification process parameters were studied.The results show that using NKA-9 resin for purification of flavonoids in Capsicum annuum,the best adsorption conditions are:Sample adsorption rate 0.59 mL/min,pH value of the sample 5 and sample volume 20mL,Capsicum annuum flavonoids adsorption rate can reach 72.03% at this condition.The optimal elution conditions are:the elution rate 1 mL/min,the ethanol solution at a concentration of 50% as the eluent and the eluent used in an amount of 50mL,Capsicum annuum
